Scientists discover process that turns normal liver cells to cancer cells

Jan. 28, 2013 ? A team of scientists from the Cancer Science Institute of Singapore (CSI) at the National University of Singapore (NUS) has identified a genetic process in which normal liver cells transform into cancerous ones. The finding provides new understanding into the pathogenesis of human cancers.

The team, which also includes researchers under the direction of Professor Guan Xin-Yuan at The University of Hong Kong, is the first to provide direct evidence that the "editing" of this protein-making sequence promotes the development of cancer.

Normally, deoxyribonucleic acid (DNA), which comprises the genetic code, serves as a template for production of ribonucleic acid (RNA) in precise fashion such that the DNA code and RNA code are identical. Editing is the process in which the RNA is changed after it is made from the DNA, resulting in production of an altered gene product.

This novel study, which was conducted from June 2010 to November 2012, was first published in the online version of Nature Medicine on 6 January 2013.

Focus on RNA

The development of liver cancer is believed to be a multi-step process, in which genetic and epigenetic alterations build up. However, it is much easier technically to detect genetic mutations through DNA sequencing. Epigenetic changes, which by definition do not involve changes in DNA sequence, have not been as well defined because it is much more difficult technically to identify them and demonstrate their role in cancer.

While recent use of next-generation sequencing of DNA and RNA have been suggestive, no apparent causal relationship between the levels of RNA editing, a process of altering RNA sequences, and cancer progression was thought to exist -- until now. As such, RNA editing is a novel epigenetic alteration in cancer.

Recognising the importance of molecular mechanisms that underlay the development of liver cancer, the research team studied the roles that RNA plays in the formation of proteins to carry out functions within the body. The researchers are the first to clearly link the RNA editing event of a protein to be significantly associated with the development of liver cancer.

Professor Daniel Tenen, Director of CSI and Principal Investigator for the study, said, "Up to now, scientists have focused on studying DNA, not RNA. Because RNA can be more easily modified than DNA, it suggests that therapeutic approaches are potentially available."

RNA Editing Process Changes Protein

In the study, the researchers performed genomic analysis of three pairs of tissue samples, each pair comprising a sample of liver cancer tissue and an adjacent non-tumour tissue, from surgical specimens of liver cancer patients.

They found that the RNA editing of a specific gene known as antizyme inhibitor 1 (AZIN1) was much higher in the tumour samples. They discovered that this editing process was catalysed by an enzyme called adenosine deaminase acting on RNA-1 (ADAR1), which changed the product of the AZIN1 protein to a form which promoted the development of cancer.

To verify their findings, the team repeated the tests on paired tissue samples of some 180 liver cancer patients.

The results obtained were consistent with the initial genomic analysis, indicating that the RNA editing of the AZIN1 gene in tumours is significantly associated with the presence of liver cirrhosis, tumor recurrence, and poorer survival rate.

Said Dr Polly Chen, Research Assistant Professor at CSI, and the lead author of the research team, "We believe that there is high possibility that the findings are applicable to other cancers, but further studies are required to ascertain this."

The Next Step

Moving forward, Prof Tenen and Dr Chen will investigate the characteristics of AZIN1 and ADAR1 to further understand their impact on tumour development. They also intend to look at these mechanisms in other cancers. They will also work on ways to correct the RNA editing process so as to block the cells' conversion from normal to malignant.

Both scientists hope that their findings will provide the medical community with a useful biomarker that can be used to detect early disorders leading to liver cancer, and even other forms of cancer, before clinical symptoms become apparent.

Kalyn's Kitchen?: Vegetarian Recipe for Green Chile and Pinto ...


This new recipe for A Month of Daily Phase One Recipes is the firstborn child of my Green Chile Mexican Casserole with Ground Beef and Layered Mexican Casserole with Chicken (that are also Phase One), and once Jake and I had the idea of trying to create a vegetarian casserole with some of those same flavors, it took us three tries to get it right.? But this final version is a definite winner, with all the flavors that make bean and green chile burritos such a favorite of many people.? Even though casseroles aren't too photogenic, I love them for the way the flavors combine to make a sum that's greater than the parts, and a leftover casserole like this in the fridge is always a nice benefit.
Dried beans are a limited food for Phase One of the South Beach Diet, but in a dish like this with other ingredients like whole green chiles, green bell pepper, and cheese, you can eat a moderate serving.? Still, I wouldn't have more than the 1/8 serving size I'm listing if you're eating this for Phase One.? And of course, this is my Meatless Monday dish for this week, and I'm betting the whole family will like this one.? Some people have told me they can't find the whole green chiles that I use to layer in these casseroles, but in Salt Lake I can find them at several stores, near the Mexican food.? If your store doesn't have them, I'd try a Mexican market.


Drain the pinto beans into a colander and rinse with cold water. In a different colander, drain the whole green chiles.

Heat the oil in a large pan, then saute onions, green peppers, minced garlic, and ground cumin.? Season with a little salt.


Add the drained pinto beans, cook about 3-4 minutes, then mash slightly. Add the green chile enchilada sauce and cook until it's mostly absorbed by the beans, then turn off heat and stir in the sour cream. One at a time, open the whole chiles with your fingers and wipe with paper towels to dry them and remove seeds.? Make into two stacks. Spray the casserole dish with non-stick spray and make a layer of half the green chiles. Then make a layer with half the bean mixture. Followed by a layer with half the cheese. Make another layer each of whole green chiles, bean mixture, and cheese. Bake with the lid on or covered with foil at 375F/190C for about 30 minutes.

Then remove the lid and bake another 20-25 minutes or until the mixture is bubbling and the top is starting to brown.? (You can put it under the broiler for a few minutes to brown more if desired.)? Serve hot.

Vegetarian Green Chile and Pinto Bean Layered Mexican Casserole
(Makes 8 servings; recipe created by Kalyn and Jake with inspiration from?Green Chile Mexican Casserole with Ground Beef and Layered Mexican Casserole with Chicken.)

Ingredients:
1 T olive oil
1 medium onion, chopped small
1/2 green bell pepper, chopped small
2 tsp. minced garlic
1 tsp. ground cumin
salt to taste
2 cans (15 oz. each) pinto beans
1 can (27 oz.) whole green chiles (Mild green chiles, not jalapenos)
1 cup Green Chile Enchilada Sauce (from a 15 oz. can; you can freeze the rest)
1/2 cup low-fat sour cream
2 cups low-fat grated cheddar cheese (I used Four Cheese Mexican Blend from Costco.)

Instructions:

Drain the pinto beans into a colander, spray with cold water until no more foam appears, and let beans drain.? In another colander drain the whole green chiles.

Heat the olive oil in a large frying pan over medium heat.? Add the onions and peppers and saute 4-5 minutes, until they are just starting to brown.? Add garlic, cumin, and salt and cook another minute or two, then add the drained beans and cook 3-4 minute, then mash the beans slightly.? (We used a potato masher, but you can use a large fork.)? Add the cup of green chile enchilada sauce and simmer until the sauce is mostly absorbed by the beans.? Then turn off the heat and stir in the sour cream.

While the bean mixture is cooking, open the whole chiles with your fingers and wipe with paper towels to dry them and remove seeds.? (This will take about 10 paper towels, but don't skip this step.)? Divide the chiles into two stacks.

In the casserole dish make a layer with half the whole green chiles, half the bean mixture, and half the cheese.? Follow with another layer each of whole green chiles, bean mixture and cheese.

Cover the casserole with the lid or with foil and bake about 30 minutes, or until cheese is all melted.? Remove the lid and bake about 20 minutes more, until casserole is bubbling and starting to brown on top.? (You can put it under the broiler for a few minutes if it seems done but you'd like the cheese more brown.)? Serve hot, with condiments like salsa, Green Tabasco Sauce, sour cream, or guacamole if desired.

This will keep in the refrigerator for several days and also freezes well.?

'The year Congress finally gets it done'

A new immigration reform proposal crafted by a bipartisan group of senators would allow for a gradual path to legalization for most of the estimated 11 million illegal immigrants currently living in the country.

A bipartisan working group of eight senators on Monday unveiled details of the proposal, calling it a "tough but fair" way to reform the nation's immigration system.

The senators' path to citizenship for illegal immigrants is less direct and could take longer than the one proposed by President Barack Obama in his immigration reform blueprint. The president will announce his push for immigration reform in more detail Tuesday in Las Vegas.

The senators' plan puts forward several hurdles before undocumented immigrants could obtain permanent residence visas, called green cards, which is the first step towards citizenship. First, immigrants would apply for temporary legal status by registering with the government, completing a background check and paying a fine. Before any immigrant could obtain a green card, "a commission comprised of governors, attorneys general, and community leaders living along the Southwest border" must declare that the government has taken enough steps to beef up border security and has also implemented a system that tracks when temporary visitors have left the country. It's unclear how long that would take, especially since border state governors are divided on how secure the border is now and ideally should be in the future.

The plan also says that no undocumented immigrant would receive a green card until every legal immigrant who is currently waiting for a green card has already received it. Wait times for green cards can be as long as 20 years in extreme cases, depending on whether the applicant is being sponsored by an employer or a family member. Family-based immigration, especially when a U.S. citizen is attempting to sponsor his or her sibling, can have especially long wait times.

The blueprint says the senators want to eliminate these wait times and drastically increase legal, high-skilled immigration, so it's possible that the bill could expedite the process. If not, illegal immigrants seeking a green card could be waiting for years.

The plan also excludes the children of illegal immigrants, commonly referred to as "Dreamers," as well as agricultural workers from the long wait for a green card.

Obama's blueprint asks for a wait time of eight years before illegal immigrants could get green cards, during which time they could live and work legally in the country. After five years with a green card, immigrants could apply to become citizens.

Speaking on the Senate floor before the announcement Majority Leader Harry Reid praised members of the committee for proposing an outline for an immigration overhaul and said he would do "everything" in his power to ensure an immigration bill passes through the upper chamber.

"Nothing short of bipartisan success is acceptable to me," Reid, a Democrat from Nevada, said."There is no reason why we should not pass comprehensive immigration reform immediately."

Immigration advocates, who generally favor a fast path to citizenship for the country's illegal immigrants, mostly praised the senators for adding to the "momentum" of reform, but did not comment on the specifics of the plan.

"Creating a 21st century immigration process won't be easy, but the framework the senators are proposing is a powerful and practical start to the legislative process, and it will make the peaks and valleys ahead much easier to traverse," said Ali Noorani, executive director of the the National Immigration Forum, in a statement.

But Chris Rickerd, policy counsel at the American Civil Liberties Union, said his organization has "concerns" that the lengthy time frame and fine might discourage people from applying to legalize. Rickerd also noted that some people might be excluded from legalization because they were prosecuted for immigration-related crimes in their states. For example in Arizona, illegal immigrants can be charged with "self smuggling," which is a felony that does not exist in other states.

Groups that favor less immigration, meanwhile, have blasted the plan as "amnesty."

"Like previous amnesty proposals, this most recent iteration creates a 'path to citizenship' for nearly all illegal aliens and offers empty promises of enforcement without providing any concrete details," said the Federation of American Immigration Reform in a statement.

The eight senators who hammered out the deal are Republicans John McCain of Arizona, Marco Rubio of Florida, Jeff Flake of Arizona and Lindsey Graham of South Carolina, and Democrats Chuck Schumer of New York, Michael Bennet of Colorado, Dick Durbin of Illinois and Robert Menendez of New Jersey.

A bipartisan group of eight senators will reveal the broad outlines of a proposal to reform U.S. immigration laws today. Articles posted on?Politico?and the New York Times this morning detailed the proposal.?

Here?s what we know so far:

It would create a pathway to citizenship

The deal includes a pathway to citizenship for the nation?s estimated 11 million undocumented immigrants. Long opposed as ?amnesty? by conservatives, this is considered the hardest but most important part of any comprehensive package.

But it would not be easy.

Undocumented immigrants would be required to undergo multiple background checks, pay a fine and back taxes, learn English and pass a civics test in order to become legal residents. Anyone with a serious criminal record would be deported.

Before they are approved, immigrants would be on a ?probationary legal status? that would allow them to live and work in the United States but not qualify for federal benefits.

But first, it calls for stricter border enforcement

The pathway to citizenship would not take effect until border security has been beefed up. That would include more drones and border guards and a supervisory commission made up of governors, community leaders and law enforcement officials from the Southwest.

The proposal also calls for an electronic verification system to allow employers to check if they are hiring undocumented immigrants.?

Another section of the proposal also calls for better tracking of people in the United States on visas through an exit system at airports and seaports.

It includes parts of the DREAM Act

The proposal would also incorporate parts of the DREAM Act, a long-stalled measure to allow people brought to the U.S. illegally as children to become citizens.?

Last year, the Obama administration decided to allow tens of thousands of people in that situation to defer deportation. This proposal would presumably build off that effort.

And it would overhaul legal immigration

The proposal would overhaul the existing legal immigration system to allow more highly skilled workers in so-called STEM fields of science, technology, engineering and math, in part by giving green cards to immigrants tho obtain advanced degrees in those fields from American universities.

Supporters of an overhaul include many in the business community in areas like Silicon Valley and standalone?bills have been successful before. These measures are considered vital to help sell other parts of a comprehensive reform bill to skeptics in Congress.

The bill would also allow more low-skill workers into the country, creating a new program for?seasonal agricultural workers.?

In recent surveys, 70 percent of Americans supported the DREAM Act provisions and 62 percent supported a pathway to citizenship. Groups lobbying for STEM immigration claim to have broad support as well.

A civil rights group opposes the employment verification.

The American Civil Liberties Union said in a statement that the framework agreed on by the senators could provide important protections for illegal immigrants who are exploited by employers and live in ?constant fear? over their immigration status.

But the ACLU took issue with the proposal to require employers to use an electronic employment-verification system, calling it ?a thinly disguised national ID requirement? that would undermine employees? privacy and lead to discrimination against those ?who look or sound ?foreign.??

Kuwait MPs target financial help for citizens

KUWAIT (Reuters) - A parliamentary committee in Kuwait proposed on Sunday that the government pay a portion of the interest on citizens' personal loans and give a 1,000 Kuwaiti dinar ($3,500) gift to each Kuwaiti without such debts, state news agency KUNA reported.

The proposal from the financial and economic affairs committee, which is made up of members of parliament, would need approval of the wider assembly and the country's ruler if it is to be passed into law.

Lawmakers elected in December had originally sought a complete bailout of billions of dollars of household debt but were met with strong resistance from policymakers who said the plans were not feasible.

If the proposal becomes law it would not be the first time that Kuwait, one of the world's richest countries per capita, gives out such financial aid.

In 2011, to mark three major anniversaries, ruler Sheikh Sabah al-Ahmad al-Sabah granted 1,000 dinars to each of the country's 1.2 million citizens and as well as free food rations for 13 months.

Kuwait's oil wealth and generous welfare state have helped to shield the Gulf country from severe Arab Spring-style unrest, although there have been frequent demonstrations over political participation and other local issues.

Under the plan proposed on Sunday, the government would pay off interest incurred on loans. KUNA said this applied to the period between January 2002 to April 2008, citing committee rapporteur Safa al-Hashem. ($1 = 0.2818 Kuwaiti dinars)

Consumer alert: new health care markets on the way

WASHINGTON (AP) -- Buying your own health insurance will never be the same.

This fall, new insurance markets called exchanges will open in each state, marking the long-awaited and much-debated debut of President Barack Obama's health care overhaul.

The goal is quality coverage for millions of uninsured people in the United States. What the reality will look like is anybody's guess ? from bureaucracy, confusion and indifference to seamless service and satisfied customers.

Exchanges will offer individuals and their families a choice of private health plans resembling what workers at major companies already get. The government will help many middle-class households pay their premiums, while low-income people will be referred to safety-net programs they might qualify for.

Most people will go online to pick a plan when open enrollment starts Oct. 1. Counselors will be available at call centers and in local communities, too. Some areas will get a storefront operation or kiosks at the mall. Translation to Spanish and other languages spoken by immigrants will be provided.

When you pick a plan, you'll no longer have to worry about getting turned down or charged more because of a medical problem. If you're a woman, you can't be charged a higher premium because of gender. Middle-aged people and those nearing retirement will get a price break: They can't be charged more than three times what younger customers pay, compared with six times or seven times today.

If all this sounds too good to be true, remember that nothing in life is free and change isn't easy.

Starting Jan. 1, 2014, when coverage takes effect in the exchanges, virtually everyone in the country will be required by law to have health insurance or face fines. The mandate is meant to get everybody paying into the insurance pool.

Obama's law is called the Affordable Care Act, but some people in the new markets might experience sticker shock over their premiums. Smokers will face a financial penalty. Younger, well-to-do people who haven't seen the need for health insurance may not be eligible for income-based assistance with their premiums.

Many people, even if they get government help, will find that health insurance still doesn't come cheaply. Monthly premiums will be less than the mortgage or rent, but maybe more than a car loan. The coverage, however, will be more robust than most individual plans currently sold.

Consider a hypothetical family of four making $60,000 and headed by a 40-year-old. They'll be eligible for a government tax credit of $7,193 toward their annual premium of $12,130. But they'd still have to pay $4,937, about 8 percent of their income, or about $410 a month.

A lower-income family would get a better deal from the government's sliding-scale subsidies.

Consider a similar four-person family making $35,000. They'd get a $10,742 tax credit toward the $12,130 annual premium. They'd have to pay $1,388, about 4 percent of their income, or about $115 a month.

The figures come from the nonpartisan Kaiser Family Foundation's online Health Reform Subsidy Calculator. But while the government assistance is called a tax credit and computed through the income tax system, the money doesn't come to you in a refund. It goes directly to insurers.

Obama's law is the biggest thing that's happened to health care since Medicare and Medicaid in the 1960s. But with open enrollment for exchange plans less than 10 months away, there's a dearth of consumer information. It's as if the consumer angle got drowned out by the political world's dispute over "Obamacare," the dismissive label coined by Republican foes.

Yet exchanges are coming to every state, even those led by staunch GOP opponents of the overhaul, such as Govs. Rick Perry of Texas and Nikki Haley of South Carolina. In their states and close to 20 others that are objecting, the exchanges will be operated by the federal government, over state opposition. Health and Human Services Secretary Kathleen Sebelius has pledged that every citizen will have access to an exchange come next Jan. 1, and few doubt her word.

But what's starting to dawn on Obama administration officials, activists, and important players in the health care industry is that the lack of consumer involvement, unless reversed, could turn the big health care launch into a dud. What if Obama cut the ribbon and nobody cared?

"The people who stand to benefit the most are the least aware of the changes that are coming," said Rachel Klein, executive director of Enroll America, a nonprofit that's trying to generate consumer enthusiasm.

"My biggest fear is that we get to Oct. 1 and people haven't heard there is help coming, and they won't benefit from it as soon as they can," she added. "I think it is a realistic fear."

Even the term "exchange" could be a stumbling block. It was invented by policy nerds. Although the law calls them "American Health Benefit Exchanges," Sebelius is starting to use the term "marketplaces" instead.

Polls underscore the concerns. A national survey last October found that only 37 percent of the uninsured said they would personally be better off because of the health care law. Twenty-three percent said they would be worse off in the Kaiser poll, while 31 percent said it would make no difference to them.

Insurers, hospitals, drug companies and other businesses that stand to benefit from the hundreds of billions of dollars the government will pump in to subsidize coverage aren't waiting for Washington to educate the public.

Blue Cross and Blue Shield plans, for example, are trying to carve out a new role for themselves as explainers of the exchanges. Somewhere around 12 million people now purchase coverage individually, but the size of the market could double or triple with the new approach, and taxpayers will underwrite it.

"Consumers are expecting their health insurance provider to be a helpful navigator to them," said Maureen Sullivan, a senior vice president for the Blues' national association. "We see 2013 as a huge year for education."

One goal is to help consumers master the "metals," the four levels of coverage that will be available through exchange plans ? bronze, silver, gold, and platinum.

Blue Cross is also working with tax preparer H&R Block, which is offering its customers a health insurance checkup at no additional charge this tax season. Returns filed this year for 2012 will be used by the government to help determine premium subsidies for 2014.

"This tax season is one of historical significance," said Meg Sutton, senior advisor for tax and health care at H&R Block. "The tax return you are filing is going to be key to determining your health care benefits on the exchange."

Only one state, Massachusetts, now has an exchange resembling what the administration wants to see around the country. With six years in business, the Health Connector enrolls about 240,000 Massachusetts residents. It was created under the health overhaul plan passed by former Republican Gov. Mitt Romney and has gotten generally positive reviews.

Connector customer Robert Schultz is a Boston area startup business consultant who got his MBA in 2008, when the economy was tanking. Yet he was able to find coverage when he graduated and hang on to his insurance through job changes since. Schultz says that's freed him to pursue his ambition of becoming a successful entrepreneur ? a job creator instead of an employee.

"It's being portrayed by opponents as being socialistic," Schultz said.. "It is only socialistic in the sense of making sure that everybody in society is covered, because the cost of making sure everybody is covered in advance is much less than the cost of putting out fires."

The Connector's executive director, Glen Shor, said his state has proven the concept works and he's confident other states can succeed on their own terms.

"There is no backing away from all the challenges associated with expanding coverage," Shor said. "We are proud in Massachusetts that we overcame what had been years of policy paralysis."

(AP) ? Gunmen killed two Iraqi soldiers and abducted three others in Fallujah on Saturday as hundreds of mourners gathered in the restive western city for the funeral of protesters killed during a shooting by army troops a day earlier, according to officials.

The attacks and kidnappings appeared to be in retaliation for the deaths of protesters in clashes Friday, and are likely to further strain tensions between Iraq's minority Sunnis and the Shiite-led central government.

Also on Saturday, Iraqi lawmakers said parliament has approved a law that would limit the terms of the prime minister, president and parliament speaker to a maximum of two terms. The measure, which must still be approved by Iraq's president, could pose a challenge to Prime Minister Nouri al-Maliki's plans to seek the post again in 2014.

Fallujah police Maj. Rasheed al-Adeeli said one of the soldiers killed was hit by a sniper on the outskirts of the city. Another was shot dead when gunmen attacked a military post where soldiers were packing up their equipment on the northern edge of the city.

Gunmen ambushed the car of three off-duty soldiers on the outskirts of Fallujah and kidnapped them, according to state television and a provincial police official reached by The Associated Press. The official, who spoke on condition of anonymity because he was not authorized to release the information, said authorities have launched a manhunt for the missing men.

Mourners hoisted caskets and waved Iraqi flags while shouting "Allahu Akbar!" or God is great. The funeral procession took place in central Fallujah, not far away from the city's cemetery.

Iraqi soldiers opened fire Friday on stone-throwing demonstrators near Fallujah, killing at least five, according to local authorities. Two other soldiers were killed Friday in apparent payback for the protesters' deaths.

Sunnis angry over perceived second-class treatment and what they see as unfair policies targeting their sect began demonstrating last month in Anbar. The protests have since spread to other predominantly Sunni areas.

The protesters are demanding the release of detainees and the cancellation of a tough counterterrorism law and other policies they believe overwhelmingly target Sunnis. Many link their cause with the broader Arab Spring and are calling for the downfall of the government altogether.

International rights group Amnesty International pressed the Iraqi government to immediately investigate the protesters' deaths and make its findings public.

"Anyone found responsible for abuses ? including anyone found to have used excessive force against protestors ? must be brought to justice," said Ann Harrison, Amnesty's deputy program director for the region. She also urged authorities to ensure security forces are properly trained and equipped to deal with demonstrators in a way that respects human rights.

Iraqi state television aired a statement from the Defense Ministry on Friday evening saying it would investigate what happened in Fallujah. Repeated efforts to reach ministry officials directly were unsuccessful.

The United Nations envoy to Iraq, Martin Kobler, said the U.N. is increasingly concerned about the rising level of tension within the country. He said he would welcome a decision by the defense ministry to investigate the incident.

"We urge the government to exert self-restraint in law and order, and respect human rights," he said in an interview. "Our message to the demonstrators is to exercise their right to demonstrate peacefully."

Kobler also called on political leaders across Iraq to "sit together in a constructive dialogue."

Muhammed al-Khalidi, a Sunni lawmaker, said that 170 lawmakers voted in favor of the law limiting the terms on the prime minister and other top officials. He said lawmakers from al-Maliki's State of Law bloc boycotted the session.

Mushriq Naji, a lawmaker from the bloc headed by firebrand cleric Moqtada al-Sadr, confirmed the vote. He said the measure is needed "to prevent the return of the dictatorship to Iraq."

"This law ensures that no person can stay in power for an unlimited time and that democracy will continue in this country," Naji said.

Putting the law into force could prove challenging, however.

Iraq's president must sign off on the legislation. The current president, Jalal Talabani, is incapacitated following a stroke and is being treated in a German hospital, and it is unlikely he would be able to sign the law into force.

Legal expert Tariq Harb downplayed the significance of the vote. Even if it were to be enacted, the supreme court could rule that it contradicts the constitution and is therefore invalid, he said.

"This vote could be part of a political game because the Iraqi constitution does not limit the terms for the prime minister and the parliament speaker," he said. "Also, the same parliament could convene at any time in the future and decide to abolish the new law if the political map changes."

Cancer drug developer ImmunoGen takes bigger loss

WALTHAM, Mass. (AP) -- Drug developer ImmunoGen Inc. said Friday its fiscal second quarter nearly doubled, as it continued developing treatments for cancer.

ImmunoGen does not have any approved drugs. Its most advanced experimental product is T-DM1, which includes the main ingredient in Roche's drug Herceptin, used mainly to treat breast cancer. The Food and Drug Administration is scheduled to make a decision on T-DM1 by Feb. 26.

ImmunoGen said it expects to report clinical trial results from its three most advanced wholly-owned drugs in 2013 and will start clinical testing of a fourth drug.

The company is developing T-DM1 through a partnership with Roche. The drug contains trastuzumab, the active ingredient in Herceptin, with a second drug and a chemical that keeps the drugs linked until they reach a cancer cell, where the cocktail can be released. ImmunoGen developed the technology that combined the drugs, and it will get royalty payments on sales if the product is approved.

The companies have asked the FDA and European regulators to approve T-DM1 as a treatment for breast cancer. Roche is running additional clinical trials of T-DM1, studying the drug in different settings as a treatment for breast cancer as well as gastric cancer. ImmunoGen is running its own trials of drugs designed to treat lung cancer and multiple myeloma, ovarian cancer, and blood cancers.

In the fiscal quarter ended Dec. 31, ImmunoGen said it lost $24.4 million, or 29 cents per share. A year ago it took a loss of $12.8 million, or 17 cents per share.

A 10-percent increase in the number of outstanding shares made the loss in the recent quarter seem smaller on a per-share basis.

Revenue fell to $2.6 million from $7.6 million, because the company received fewer payments from its drug development partners.

Analysts were expecting a loss of 25 cents per share and $5.2 million in revenue, according to FactSet.

Shares of ImmunoGen lost 48 cents, or 3.2 percent, to $14.87 in midday trading.

Death toll rising in Egypt riot after soccer violence verdict

CAIRO (AP) ? An Egyptian court sentenced 21 people to death Saturday on charges related to one of the world's deadliest incidents of soccer violence, touching off an attempted jailbreak and a riot that killed 16 in the Mediterranean port city that is home to most of the defendants.

The verdict follows deadly clashes between police and demonstrators on Friday, the second anniversary of the uprising that overthrew longtime leader Hosni Mubarak. Such cycles of violence, often lasting for weeks and costing dozens of lives, have occurred regularly over the past two years.

Die-hard soccer fans from both teams, known as Ultras, hold the police at least partially responsible for the Port Said deaths and criticize Egypt's President Mohammed Morsi for doing little to reform the force.

Al-Ahly Ultras in particular have been at the forefront of protests. But anger also is boiling in Port Said, where residents say they have been unfairly scapegoated.

Immediately after the verdict, two police were shot dead outside Port Said's main prison when angry relatives tried to storm the facility to free the defendants. Police fired tear gas and rubber bullets, as well as live rounds, at the crowd outside the prison, killing 14, security officials said. Hundreds were wounded. They spoke on condition of anonymity in line with regulations.

Security officials said the military is being deployed to Port Said ? the second such deployment in less than 24 hours. The army was widely used to keep order by top generals who took over after Mubarak, but the military has kept a much lower profile since Morsi was elected as president in June.

Morsi cancelled a scheduled trip to Ethiopia Saturday and instead met for the first time with top generals as part of the newly-formed National Defense Council.

The military was also deployed overnight in the city of Suez after eight people died in clashes between security forces and protesters opposed to Morsi. Another protester was killed in Ismailiya, and security officials told the state news agency MENA that two policemen were killed in Friday's protests, bringing the death toll on the second anniversary of Egypt's uprising to 11.

Judge Sobhi Abdel-Maguid read out the death sentences related to the Feb. 1 riot in Port Said that killed 74 fans of the Cairo-based Al-Ahly team. Defendants' lawyers said all those sentenced were fans of the Port Said team, Al-Masry. Executions in Egypt are usually carried out by hanging.

The judge Saturday said in his statement read live on state TV that he would announce the verdict for the remaining 52 defendants on March 9.

Among those on trial are nine security officials, but none were handed sentences Saturday, lawyers and security officials say.

Fans of al-Ahly, whose stands were attacked by rival club Al-Masry in the Feb. 1 incident in Port Said, had promised more violence if the accused did not receive death sentences. In the days leading up to the verdict, Al-Ahly fans warned of bloodshed and "retribution". Hundreds of Al-Ahly fans gathered outside the Cairo sports club in anticipation of the verdict, chanting against the police and the government.

Before the judge could read out the names of the 21, families erupted in screams of "Allahu Akbar!" Arabic for God is great, with their hands in the air and waving pictures of the deceased. One man fainted while others hugged one another. The judge smacked the bench several times to try and contain reaction in the courtroom.

"This was necessary," said Nour al-Sabah, whose 17 year-old son Ahmed Zakaria died in the melee. "Now I want to see the guys when they are executed with my own eyes, just as they saw the murder of my son."

The verdict is not expected to calm tensions between the two rival teams. The judge is expected to make public his reasons for the death sentences March 9, when the remaining 52 defendants receive their sentences.

A Port Said resident and lawyer of one of the defendants given a death sentence said the verdict was nothing more than "a political decision to calm the public."

"There is nothing to say these people did anything and we don't understand what this verdict is based on," Mohammed al-Daw told The Associated Press by telephone.

"Our situation in Port Said is very grave because kids were taken from their homes for wearing green T-shirts," he said, referring to the Al-Masry team color.

The violence began after the Port Said's home team won the match, 3-1. Al-Masry fans stormed the pitch after the game ended, attacking Cairo's Al-Ahly fans.

Authorities shut off the stadium lights, plunging it into darkness. In the exit corridor, the fleeing crowd pressed against a chained gate until it broke open. Many were crushed under the crowd of people trying to flee.

Survivors of the riot described a nightmarish scene in the stadium. Police stood by doing nothing, they said, as fans of Al-Masry attacked supporters of the top Cairo club stabbing them and throwing them off bleachers.

Al-Ahly survivors said supporters of Al-Masry carved the words "Port Said" into their bodies and undressed them while beating them with iron bars.

While there has long been bad blood between the two rival teams, many blamed police for failing to perform usual searches for weapons at the stadium.

Both Al-Ahly Ultras and Al-Masry Ultras widely believe that ex-members of the ousted regime of Hosni Mubarak helped instigate the attack, and that the police at the very least were responsible for gross negligence. It is not clear what kind of evidence, if any, was presented to the court to back up claims that the attack had been orchestrated by regime officials.

"The police are thugs!" yelled relatives of the deceased inside the courtroom before the judge took the bench.

As is customary in Egypt, the death sentences will be sent to the nation's top religious authority, the Grand Mufti, for approval, though the court has final say on the matter.

All of the defendants ? who were not present in the courtroom Saturday for security reasons ? have the right to appeal the verdict.

The melee was the world's deadliest soccer violence in 15 years.

The Ultras are proud of their hatred for the police, who were the backbone of Mubarak's authoritarian rule. They then then directed their chants against the military rulers who took over after Mubarak's ouster.

Ultras from several Egyptian sports clubs were engaged in deadly clashes with police near the Interior Ministry headquarters in Cairo that killed 42 people less than three months before the soccer melee in Port Said.

Lawyer: No proof to charge Romanians for art theft

BUCHAREST, Romania (AP) ? A lawyer for three Romanians accused of stealing valuable paintings from a museum in the Netherlands insisted Friday there was insufficient evidence to charge them.

At a court hearing in Bucharest, the Romanian capital, defense lawyer Doina Lupu urged that the three men not be extradited to the Netherlands for allegedly stealing seven works by Picasso, Monet and Matisse, among others.

"There is no evidence that this was an organized criminal group," she said. "The arrests were based on assumptions and on simple declarations and these are not enough. "

All three men deny any links to the Oct. 16 theft at Rotterdam's Kunsthal gallery.

A prosecutor told the court Friday that suspect Radu Dogaru took two paintings ? one each by Henri Matisse and Paul Gauguin ? to be evaluated by Maria Dragu, the curator for foreign paintings at the Romanian National Art Museum.

Dogaru denied this and Dragu is on leave, the Romanian museum said Friday.

The other two suspects, Alexandru Bitu and Eugen Darie, also denied being involved in the theft. Darie did tell the court he had visited the Kunsthal gallery twice to look at its bronze sculptures.

The Bucharest Court ruled Friday that the men should be kept in custody pending an investigation.

The thieves broke in Oct. 16 through a rear emergency exit at the gallery, grabbed the paintings off the wall and fled, all within two minutes.

The stolen paintings came from the private Triton Foundation, a collection of avant-garde art put together by multimillionaire Willem Cordia, an investor and businessman, and his wife, Marijke Cordia-Van der Laan. Willem Cordia died in 2011.

The stolen paintings were: Pablo Picasso's 1971 "Harlequin Head;" Claude Monet's 1901 "Waterloo Bridge, London" and "Charing Cross Bridge, London;" Matisse's 1919 "Reading Girl in White and Yellow;" Gauguin's 1898 "Girl in Front of Open Window;" Meyer de Haan's "Self-Portrait," around 1890; and Lucian Freud's 2002 work "Woman with Eyes Closed."

The gallery said it had a "state-of-the-art" automated alarm system.

Head Coach Tom Moore Picks Up Career Win No. 100; Men's ...

HAMDEN, Conn. ? Quinnipiac University men's basketball head coach Tom Moore picked up his 100th career win on Thursday night, as the Bobcats made it eight straight wins over Central Connecticut with an 85-78 victory at the TD Bank Sports Center. Moore became the fastest coach to win 100 in the Division I era and second fastest coach all-time in program history.

Shaq Shannon (Philadelphia, Pa.) was the difference-maker for the Bobcats ? as the junior guard poured in 18 points which included six from long-range. He was the first player to hit six threes since James Johnson accomplished the feat back in the 2010-11 season with seven against Bryant. Ike Azotam (Boston, Mass.) added 13 points and grabbed a game-high 17 rebounds, while fellow front court mate Jamee Jackson (Newark, N.J.) chipped in with 14 points and eight rebounds. Zaid Hearst (Bethesda, Md.) finished with 17 points making it five straight with at least 10 points. Dave Johnson (Jackson, N.J.) composed a steady offense with eight assists, four points and three rebounds.

The game didn't see more than a six-point separation through the first half and most of the second. However, trailing by one point with 13:08 showing on the clock, Quinnipiac (7-12, 3-4 NEC)?opened up its largest lead of the game with a 16-6 run behind three, threes, two from Shannon, and several easy buckets underneath. The Blue Devils would cut their deficit to three points in the waning minutes, but clutch free-throw shooting in the closing seconds put the game out of reach en route to the historic win.

The two teams matched up very evenly throughout the game; but it was the discrepancy on the boards that granted Quinnipiac the edge it needed. The Bobcats garnered a plus-20 margin on the glass, 51-31 - which also included 17 on offensive end and 15 second chance points. Both teams shot 47 percent from the floor and only combined for 14 turnovers.

Kyle Vinales, the NEC's leading scorer, led the Blue Devils with a game-high 32 points on 12-of-20 shooting. Matt Hunter added 13, while Joe Efese and Malcolm McMillan each had 10.

In a series that dates back to the 1956-57 campaign, wins are beginning to pile up for Quinnipiac. Over the last five seasons, QU has won nine out of the last 10 games and is now riding a seven-game winning streak.

Prior to tip-off, the Quinnipiac University and Central Connecticut State University Departments of Athletics and Recreation recognized three-time National Champion and former UConn men's basketball head coach Jim Calhoun for his leadership in bringing the entire state of Connecticut the highest level of recognition and success in Division I men's basketball. Quinnipiac and CCSU presented Coach Calhoun with a donation that will go directly to the Jim and Pat Calhoun Cardiology Center.

The Bobcats will wrap up the fourth week of NEC play on Saturday afternoon against Fairleigh Dickinson. Tip-off is set for 12 p.m. with all the action televised live on MSG and FCS.

How trained literacy coaches can improve student reading comprehension

Jan. 25, 2013 ? The language and reading comprehension skills of low-income upper elementary-school students -- especially English-language learners -- can improve markedly if trained literacy coaches engage teachers in conducting interactive text discussions with students, according to a three-year University of Pittsburgh study.

The Pitt researchers report in the journal Learning and Instruction that language and reading comprehension showed measurable improvement for young students when their teachers had worked "at-elbow" with content-specific literacy coaches to foster a more interactive learning environment during class reading assignments.

In the study -- one of the first of its kind -- the coaches were trained using a professional development system designed at Pitt's Institute for Learning called the Content-Focused Coaching Model? that has coaches provide teachers with the tools they need to implement rigorous, standards-based lessons. Teachers can then use the knowledge they've gained to train other teachers in their schools.

"Our goal was to create a method for closing the literacy gap between more privileged and low-income students," said study principal investigator Lindsay Clare Matsumura, a research scientists in Pitt's Learning Research and Development Center and an associate professor in Pitt's School of Education. "We found that a well-structured and content-specific approach to literacy coaching shows strong evidence of being able to really make an impact on classroom text discussion and reading achievement in these upper elementary grades -- a critical time for students to develop their higher-level reading skills."

This content-specific method, developed at Pitt's Institute for Learning within the University's Learning Research and Development Center, also lends itself to adherence of the Common Core State Standards, a national campaign aiming to increase the quality of the country's education system.

"Changing discussion patterns in classrooms is a big undertaking," said Donna DiPrima Bickel, a fellow in Pitt's Institute for Learning and codeveloper and leader of the Content-Focused Coaching Model?. "The Common Core State Standards require a broader and deeper level of comprehension from students at all grade levels beyond first grade. It's imperative that teachers learn ways of supporting students to interact effectively with a range of different types of texts. Teachers engaged with coaches trained in the Content-Focused Coaching Model? valued the support they received in helping them learn to facilitate text discussion on rigorous texts."

In a group-randomized trial, Matsumura -- along with Helen Garnier, a consultant with Pitt's Learning Research and Development Center, and Jessaca Spybrook of Western Michigan University -- investigated the effects of the Content-Focused Coaching Model?, zeroing in on the quality of text discussions in the classroom.

The researchers worked with 29 schools in a Texas school district that serves a high percentage of low-income and English-language-learning students. Half of the schools adopted the Content-Focused Coaching Model?, which entailed highly trained coaches entering schools and providing professional development training to upper elementary school teachers. The other half continued in their previously assigned literacy plan, which included the involvement of literacy coaches untrained in the coaching model.

"Many literacy coaches don't receive a lot of training on how to work effectively with teachers," said Matsumura. "So our goal was to work intensively with these coaches by teaching them how to model instructional strategies and work with teachers to better plan reading lessons. We provided them with effective strategies to share with teachers so they could boost their engagement with students."

During the study's first year, Pitt researchers collected baseline data on the quality of classroom discussions, teachers' participation in professional development, and students' reading achievement. Soon after, the coaches were placed in schools and began working with teachers on "Questioning the Author," one approach to the Content-Focused Coaching Model? in which students answer critical questions about the author and text. "Questioning the Author" was developed at Pitt's Learning Research and Development Center by Emeritus Professor Isabel Beck and Research Scientist Margaret McKeown.

Under this literacy approach, students are asked to stop throughout the reading of a book and answer thought-provoking questions. If a text is written unclearly, said Matsumura, the teacher will pause to make sure students understand what is happening and also review any unknown vocabulary. The approach is meant to result in more interactive discussions leading to better comprehension and retention, and it entails "quite a bit of planning" on the part of teachers for it to be effective, Matsumura said.

In the Pitt study, the coaches began by leading a classroom discussion while teachers observed. After several months of observation, the teachers adopted the technique into their classroom for the rest of the study. Students' reading scores were evaluated through a series of tests throughout the three years.

The team found that schools participating in the coaching intervention had a positive effect on students' reading achievement -- specifically for English-language learners, who made up 40 percent of the study's sample. English-language learners with trained teachers scored .48 of a standard deviation higher on the state reading rest than those in the comparison schools. A standard deviation is the average distance between any score in a distribution and the mean of the distribution.

"One of our suppositions is that in having these kinds of interactive discussions, you're really getting kids to talk and learn to use new vocabulary actively," said Matsumura. "Nevertheless, our study highlights the need for usage of literacy-coaching programs -- like the Content-Focused Coaching Model? -- to promote student reading achievement."

In addition to improving students' literacy at an individual level, the study had a larger, across-the-board success.

"Anyone can provide you with an anecdote about how one coach has helped one teacher but the real story here is in the systematic results," said Bickel. "Lindsay and her team retrieved data that clearly show that schools with coaches trained in the Content-Focused Coaching Model? improved literacy skills far beyond those where teachers worked with coaches who were not trained in this method. And, as a result of this study, we are able to better describe the components of an effective coaching program."

As is the case with most large-scale projects, widespread implementation of these interventions may prove to be a challenge. Therefore, Matsumura and her colleagues are now working towards delivering this coaching online.

Quoting Tim Cook: 5 Gadget-Related Sound Bites from Apple?s Latest Earnings Call

Apple held its latest earnings call yesterday ? you can read about the financial implications over in our Business & Money section ? which ended with CEO Tim Cook and CFO Peter Oppenheimer fielding questions from industry analysts.

If you?re more interested in gadgets than numbers (I know I am), the Q&A segments of Apple?s earnings calls are where most of the good stuff lives.

Here are five quotes from Tim Cook that relate to Apple?s hardware lines.

Android phones with 5-inch and larger screens are getting more and more popular, but we probably shouldn?t expect a huge iPhone anytime soon:

The iPhone 5 offers ? as you know ? a new 4-inch Retina display, which is the most advanced display in the industry. No one comes close to matching the level of quality as the Retina display. It also provides a larger screen size for iPhone customers without sacrificing the one-handed ease of use that our customers love. So we put a lot of thinking into screen size and believe we picked the right one.

Why Apple rumors are tricky (likely referring to recent reports of iPad screen production slowdowns):

I know there?s been lots of rumors about order cuts and so forth, and so let me just take a moment to make a comment on these. I don?t want to comment on any particular rumor because I would spend my life doing that, but I would suggest it?s good to question the accuracy of any kind of rumor about build plans, and also stress that even if a particular data point were factual, it would be impossible to accurately interpret the data point as to what it meant for our overall business, because the supply chain is very complex and we obviously have multiple sources for things, yields might vary, supplier performance can vary, the beginning inventory positions can vary. I mean, there?s just an inordinate long list of things that would make any single data point not a great proxy for what?s going on.

Why Apple won?t produce a bunch of different versions of the iPhone to try to hold (or win back) smartphone market share:

The most important thing to Apple is to make the best products in the world that enrich customers? lives. That?s our high-order bid. That means that we aren?t interested in revenue for revenue?s sake. We could put the Apple brand on a lot of things and sell a lot more stuff, but that?s not what we?re here for. We want to make only the best products. And so what does that mean for market share? We?ve been able to do that, and I think we?ve had a great track record here on iPod of doing different products at different price points and getting a reasonable share from doing that. I wouldn?t view these things as mutually exclusive as some might. But the high-order bid is making a great product that enriches customers? lives, and so that?s what we?re focused on.

On the subject of Apple TV (the $99 box) versus an Apple TV (the mythical TV set), and the difficulty of getting content providers to play ball:

In terms of the product that we sell today ? the Apple TV ? we sold more last quarter than we?ve ever sold before, eclipsing two million during the quarter. It was up almost 60% year-on-year and so there is actually very, very good growth in that product. What was a small niche at one time of people that loved it is a much larger number that love it. I have said in the past this is an area of intense interest for us, and it remains that. I tend to believe that there is a lot we can contribute in this space. And so we continue to pull the string and see where it leads us, but I don?t want to be more specific.

Why the iPhone eating into iPod sales is no big deal. Same for the iPad eating into Mac sales:

In terms of cannibalization and how we think about this, I see cannibalization as a huge opportunity for us. One, our base philosophy is to never fear cannibalization. If we do, somebody else will just cannibalize it, and so we never fear it. We know that iPhone has cannibalized the iPod business ? it doesn?t worry us that it?s done that. We know that iPad will cannibalize some Macs ? that doesn?t worry us. On iPad in particular, we have the mother of all opportunities here, because the Windows market is much, much larger than the Mac market is, and I think it is clear that it?s already cannibalizing some and I think there?s a tremendous amount more opportunity there. As you know, I?ve said for two or three ? actually three years now, I believe ? that I believe the tablet market would be larger than the PC market at some point, and I still believe that. And you can see by the growth in tablets and the pressure on PCs that those lines are beginning to converge.

I think the other thing for us ? maybe not for others, but for us ?is that if somebody buys an iPad Mini or an iPad and it?s their first Apple product, we have great experience through the years of knowing that when somebody buys their first Apple product, that a percentage of these people wind up buying another type of Apple product. And so, if you remember what we had termed the halo effect for some time with the iPod and with the Mac, we?re very confident that that will happen, and we?re seeing some evidence of that on the iPad as well. And so I see cannibalization as a huge opportunity.
